It is important to optimize the condition of in vitro maturation (IVM) on porcine oocytes. Allicin that exhibits strong antioxidant activity prevents proper oocyte functions from reactive oxygen species (ROS). This study investigated to confirm effect of allicin treatment on porcine oocyte maturation and its developmental capacity. The porcine oocytes were cultured in media supplemented with 0, 0.01, 0.1, 1, 10 and 100 μM allicin (0 AL, 0.01 AL, 0.1 AL, 1 AL, 10 AL and 100 AL) during IVM. The 0.1 AL treated group was significantly increased in normal Mrate compared to in control group (*p < 0.1). The level of ROS was not different among the control and AL treated groups at Mstage. Similarly, there were not different between control and 0.1 AL treatment group during the each maturation stage (GV, GVBD, Mand M) of oocyte. After parthenogenetic activation, rate of cleavage was significantly increased in 0.1 AL treatment group compared to control (**p < 0.05). Although the total cell number was not different in all groups, proportion of blastocyst was significantly increased in 0.1 AL treated group. The expression of anti-apoptosis related genes had considerably increased in 0.1 AL treated group compared to in other groups. These results indicate that maturation medium supplemented with allicin during IVM can influence on oocyte maturation, and improve subsequent development of embryo via decreasing expression of anti-apoptosis related genes.
