Introduction
Sperm treatment methods affect sperm
fertility and embryo developmental potential in in vitro fertilization (IVF) system and selection of high quality
sperm for production of elite Jeju Black Cattle (JBC) zygotes is necessary. In
this study, we selected an effective sperm treatment method and superior
breeding bull semen.
Materials
and Methods
In order to find out the most suitable
sperm treatment method, the following methods of percoll gradient, triladyl and
percoll-triladyl combination (SF, T and TSF) were used to IVF. To confirm the
capacitation, sperm was stained with chlorotetracycline (CTC). We checked the
capacity of JBC sperm A, B and C through embryo development after IVF with
them.
Results
and Discussion
The blastocyst formation rate was increased
in T group than SF and TSF groups. The T method can collect only a small amount
of sperm, and the SF method is toxic. To compensate for this disadvantage, we
combined the T and SF methods and then experimented with the TSF method. As the
result of CTC staining, acrosome-reacted sperm (AR pattern) is the highest in sperm
C. After IVF, the blastocyst formation rate was the highest in sperm C used
group, but the total cell number has no difference. These results demonstrate
that sperm C was effective in production of elite IVF embryos.
